Endocrinology | Results of Phase III Trial (GLORY-2) Published in JAMA

Results from the Phase III trial (GLORY-2) evaluating mazdutide for weight reduction in Chinese adults with obesity, a novel dual glucagon (GCG)/glucagon-like peptide-1 (GLP-1) receptor agonist, were presented at the 86th Scientific Sessions of the American Diabetes Association (ADA) and simultaneously published in the Journal of the American Medical Association.

Led by Professor Ji Linong of Peking University People’s Hospital (PKUPH), the study provided important evidence on the efficacy and safety of mazdutide 9 mg as a weight management option for Chinese patients with moderate or severe obesity.

The prevalence of obesity in China is high, and people with a BMI exceeding 30 kg/m² often have a heavier cardiometabolic burden which requires special attention in clinical weight management. There remains an unmet clinical need for pharmacological treatment in this population. The findings from GLORY-2 trial may offer a new therapeutic option for individuals with moderate or severe obesity. The randomized, double-blind, placebo-controlled trial enrolled 462 participants, including 16% with type 2 diabetes, who received either mazdutide 9 mg or placebo over 60 weeks. Participants had a mean baseline weight of 94.0 kg and a mean BMI of 34.3 kg/m².

At week 60, patients treated with mazdutide 9mg achieved a mean weight reduction of 18.55%, compared with 3.02% in the placebo group. Notably, 44.0% of participants in the mazdutide 9mg group achieved weight reduction of at least 20% from baseline vs. 2.6% in the placebo group. Weight demonstrated a persistent reduction throughout the study, with no clear plateau observed. Mazdutide also improved multiple cardiometabolic risk factors, including waist circumference, blood pressure, lipid profile, and uric acid levels, while maintaining a favorable safety profile.


Following the publication of the GLORY-1 trial (Once-Weekly Mazdutide in Chinese Adults with Obesity or Overweight) in The New England Journal of Medicine in 2025, the findings in GLORY-2 trial further highlighted Chinese investigators’ growing contributions to the research and treatment of obesity and metabolic disease in the world.
